does it matter where the pistons are....... ie one tdc , or should they both be in middle of stroke...... or it dont matter....

doesnt matter as the cyclinderis sealed from either side.

Its easy, plug the holes, pressure to 5 or 6 psi and come back and check it after a bit, say 5. 15 or 30mins. see what its lost, hopefully nothing. YPVS do leak a bit but on an LC you prob wont lose much at all.
If youve lost pressure, grab some soapy water (I use window cleaner as it evaporates and is very bubbly) and spray it over the carb rubbers first, chances are thats where its leaking. Fix each leak and try again.

Personally I've been dicking about/tuning 2 stroke bikes since '66. Even fielded TZ's for Yama in early 70's... albeit in a previous life path. Never actually done a leak down test. Never had motivation nor need to.

IMO the leak down test on 2T is to check air - compression leaking (seals, gaskets ..) or false air suction.
That is a completely different thing than pure compression test (piston vs rings vs cylinder vs head gasket).
So if you think that you engine is on low compression, check that first.
