Coming to the end of a refurb on a freinds RZ250.. As regards the top end should i hand tighten the barrels, then torque the head, then fully tighten the barrel nuts ( as in KR1S )..or fully tighten barrels down then the head..? Tks..

I always do head in 2 stages first then base nuts

Remember to re-torque after a few heat cycles, and even several times until the barrels have settled and the torque is correct.
I also favour the first heat cycle without coolant in so it beds defore any fluid is around.

Jon Hi Jon, what's your preferred method of torque process after a heat cycle, straight on with the wrench and nip up to required value or back of the bolt 1/8 of a turn and then re-torque to required value?

I wouldn't loosen them first.
I would however do it in stages. Set the torque wrench slightly below the required figure and torque in sequence. Then repeat at the set value.
